The Derby runner-up, trained by David Lanigan, has the Prix Niel at Longchamp the following day as alternative engagement should there be a late change of plan.
"He worked very nicely yesterday and he's on course to go to Doncaster for the St Leger all being well," said Alan Cooper, racing manager to owners the Niarchos Family.
"He's a very good horse and the plan is to run there and the Prix Niel is there in case we need it.
"The Leger is the objective and the Classic over one mile and six furlongs is about staying.
"The horse is very well and David is pleased with him."
Since chasing home Camelot in the Derby, Main Sequence has been an unlucky fourth in the Grand Prix de Paris at Longchamp and a strong-finishing second in the Great Voltigeur Stakes behind Thought Worthy at York.
Main Sequence can be backed at 11.5n/a for the St Leger.
